The spring barley (Hordeum vulgare L.) cultivar Golden Promise was modified using CRISPR/Cas-mediated genome editing to generate the HvPIL1 -2 line with reduced plant height. The modification targets the endogenous HvPIL1 (phytochrome interacting factor-like 1) gene, which is involved in the regulation of stem growth. Homozygous HvPIL1 -2 plants exhibited a reduction in height of approximately 10 cm under greenhouse conditions. The reduced stature may contribute to decreased susceptibility to lodging.

Information on the modified DNA sequences
The HvPIL1 -2 line contains a two-base-pair deletion of adenine and cytosine that disrupts the reading frame, resulting in a non-functional HvPIL1 protein. HvPIL1 encodes a basic helix-loop-helix (bHLH) transcription factor involved in the regulation of stem growth. The gene is light responsive and contributes to the activation of genes involved in cell wall expansion. Its expression fluctuates throughout the day, peaking around midday, and is downregulated under drought conditions. Homozygous HvPIL1 -2 mutant plants exhibited a reduction in height of approximately 10 cm when grown under greenhouse conditions.
